Ikaros, Notch, and GATA1 cross paths during megakaryopoiesis ----------------------------------------------------------------------------------------------------Ivan Maillard UNIVERSITY OF MICHIGAN In this issue of Blood, Malinge et al describe new reciprocal interactions between Ikaros, Notch, and GATA transcription factors during megakaryocyte development. Ikaros represses megakaryocytic genes and selected Notch targets, while being turned off by GATA1 upon terminal differentiation.

Genetic analysis of hierarchical regulation for Gata1 and NF-E2 p45 gene expression in megakaryopoiesis.
GATA1 and NF-E2 p45 are two important regulators of megakaryopoiesis. Whereas GATA1 is known to regulate the p45 gene, details of the GATA1 contribution to the spatiotemporal expression of the p45 gene remain to be elucidated. To clarify the relationship between GATA1 and p45, we performed genetic complementation rescue analysis of p45 function in megakaryocytes utilizing the hematopoietic regu...
متن کاملNotch activation is an early and critical event during T-Cell leukemogenesis in Ikaros-deficient mice.
The Ikaros transcription factor is both a key regulator of lymphocyte differentiation and a tumor suppressor in T lymphocytes. Mice carrying a hypomorphic mutation (Ik(L/L)) in the Ikaros gene all develop thymic lymphomas. Ik(L/L) tumors always exhibit strong activation of the Notch pathway, which is required for tumor cell proliferation in vitro. Notch activation occurs early in tumorigenesis ...
متن کاملIkaros represses the transcriptional response to Notch signaling in T-cell development.
Notch activity is essential for early T-cell differentiation, but aberrant activity induces T-cell transformation. Thus, Notch target genes must be efficiently silenced in cells where Notch activity is no longer required. How these genes are repressed remains poorly understood. We report here that the Ikaros transcription factor plays a crucial role in repressing the transcriptional response to...
متن کاملInduction of hyperproliferative fetal megakaryopoiesis by an N-terminally truncated GATA1 mutant.
Two GATA1-related leukemias have been described: one is an erythroleukemia that develops in mice as a consequence of diminished expression of wild-type GATA1, whereas the other is an acute megakaryoblastic leukemia (AMKL) that arises in Down syndrome children as a consequence of somatic N-terminal truncation (DeltaNT) of GATA1. We discovered that mice expressing the shortened GATA1 protein (Del...
متن کاملPACAP and its receptor VPAC1 regulate megakaryocyte maturation: therapeutic implications.
Megakaryocytes and platelets express the Gs-coupled VPAC1 receptor, for which the pituitary adenylyl cyclase-activating peptide (PACAP) and the vasointestinal peptide (VIP) are agonists. We here demonstrate a regulatory role for VPAC1 signaling during megakaryopoiesis. A total of 2 patients with trisomy 18p with PACAP overexpression and transgenic mice overexpressing PACAP in megakaryocytes hav...
